A cell-permeable, rhodanine-acetic acid analog that chelates the active site Zn2+ and acts as a potent inhibitor of anthrax lethal factor (LF) metalloproteinase (Ki = 32 nM). Has only a minimal inhibitory effect on MMP-2 and MMP-9. Shown to effectively prevent MAPKK1 proteolysis (< 2 µM) by LF and offer protection against LF and protective antigen (PA) cytotoxic effects in RAW264.7 cells (IC50 < 5 µM). BI-11B3 has been shown to double the survival rates of mice challenged with anthrax spores and treated with ciproflaxin.
