pET Expression Systems and pET Expression Systems plus Competent Cells provide core reagents needed for target gene cloning and expression.
Components: pET Expression Systems
Components for pET Expression Systems are similar for all systems unless otherwise stated with the specific pET Expression System description. pET Expression Systems include:
• 10 µg pET vector DNA (for each indicated plasmid)
• 0.2 ml BL21 Glycerol Stock
• 0.2 ml BL21(DE3) Glycerol Stock
• 0.2 ml BL21(DE3)pLysS Glycerol Stock
• 0.2 ml Induction Control Glycerol Stock
Components: pET Expression Systems plus Competent Cells
pET Expression Systems plus Competent Cells contain all of the components of the specific pET Expression System, as well as the following additional components, unless otherwise stated with the specific pET Expression System description:
• 0.2 ml NovaBlue Competent Cells
• 0.2 ml BL21(DE3) Competent Cells
• 0.2 ml BL21(DE3)pLysS Competent Cells
• 2 × 0.2 ml SOC Medium
• 10 µl Test Plasmid
These components are sufficient for up to 10 transformations in each host.

pET Expression System 33b plus Competent Cells
The pET-33b(+) vector is a unique expression plasmid that encodes a 5-aa recognition sequence for the catalytic subunit of cAMP-dependent protein kinase (protein kinase A; PKA) from heart muscle (Blanar 1992, de Arruda 1995). The vector-encoded fusion sequences available on pET-33b(+) include His•Tag®, thrombin cleavage site, PKA recognition sequence (ArgArgAlaSerVal), and T7•Tag® peptides. Fusion proteins expressed from this plasmid can be affinity purified with either His•Bind® resins or T7•Tag Antibody Agarose, and then labeled in vitro in a simple reaction with 32P-γ-ATP and purified PKA (available in the PKAce™ Kit). Proteins can be labeled to at least 105 cpm/µg in 10 minutes, and can be used directly as sensitive and specific probes for protein:protein interactions (de Arruda 1995). A powerful method that uses such probes for rapid mapping of interacting protein domains has been described (Burgess 1998, Arthur 1998).
