BLT5615 is a host strain for the growth of the bacteriophage T7Select10-3, T7Select1-1 and T7Select1-2, which require an exogenous source of capsid protein. BLT5615 carries an ampicillin-resistant plasmid that supplies large amounts of capsid protein. BLT5615 is a less inhibitory host than
BLT5403, where 10A expression is controlled by a lacUV5 promoter. Phage displaying larger proteins (600 amino acids) that gave tiny plaques and accumulated deletions in the capsid fusion gene on BLT5403, gave larger plaques and produced lysates without detectable deletion phage on BLT5615. BLT5615 may also be a useful host for making T7Select10-3, T7Select1-1 and 1-2 libraries. Growth of T7Select10-3, T7Select1-1 and 1-2 clones on BLT5615 requires the addition of IPTG to induce production of the 10A protein.
